    Expectation propagation as a solution for digital communication systems.

    In the context of digital communications, a digital receiver is required to provide an estimation of the transmitted symbols. Nowadays channel decoders highly benefit from soft (probabilistic) estimates for each transmitted symbol rather than from hard decisions. For this reason, digital receivers must be designed to provide the probability that each possible symbol was transmitted based on the received corrupted signal. Since exact inference might be unfeasible in terms of complexity for high-order scenarios, it is necessary to resort to approximate inference, such as the linear minimum mean square error (LMMSE) criterion. The LMMSE approximates the discrete prior information of the transmitted symbols with a Gaussian distribution, which causes a degradation in its performance. In this thesis, an alternative approximate statistical technique is applied to the design of a digital probabilistic receiver in digital communications. Specifically, the expectation propagation (EP) algorithm is investigated to find the Gaussian posterior probability density function (pdf) that minimizes the Kullback-Leibler (KL) divergence with respect to the true posterior pdf. Two different communication system scenarios are studied: a single-input singleoutput (SISO) digital communication system with memory channel and a multipleinput multiple-output (MIMO) system with memoryless channel. In the SISO scenario, three different designs of a soft standalone and turbo equalizer based on the EP algorithm are developed: the block or batch approach, the filter-type version that emulates theWiener filter behavior and the smoothing equalizer which proceeds similarly to a Kalman smoother. Finally, the block EP implementation is also adapted to MIMO scenarios with feedback from the decoder. In both scenarios, the EP is applied iteratively, including a damping mechanism and a control to avoid negative values of variances, which would lead to instabilities (specially for high-order constellations). Experimental results included through the thesis show that the EP algorithm applied to communication systems greatly improves the performance of previous approaches found in the literature with a complexity slightly increased but still proportional to that of the LMMSE. These results also show the robustness of the algorithm even for high-order modulations, large memory channels and high number of antennas. Los decodificadores de canal actuales se benefician enormemente de estimaciones suaves (probabilísticas) de cada símbolo transmitido, en vez de estimaciones duras. Por este motivo, los receptores digitales deben diseñarse para proporcionar la probabilidad de cada posible símbolo que fue transmitido en base a la señal recibida y corrupta. Dado que la inferencia exacta puede no ser posible en términos de complejidad para escenarios de alto orden, es necesario recurrir a inferencia aproximada, como por ejemplo el criterio de linear minimum-mean-square-error (LMMSE). El LMMSE aproxima la información a priori discreta de los símbolos transmitidos con una distribución Gaussiana, lo cual provoca una degradación en su resultado. En esta tesis, se aplica una técnica alternativa de inferencia estadística para diseñar un receptor digital probabilístico de comunicaciones digitales. En concreto, se investiga el algoritmo expectation propagation (EP) con el objetivo de encontrar la función densidad de probabilidad (pdf) a posteriori Gaussiana que minimiza la divergencia de Kullback-Leibler (KL) con respecto a la pdf a posteriori verdadera. Se estudian dos escenarios de comunicaciones digitales diferentes: un sistema de comunicaciones single-input single-output (SISO) con canales con memoria y un sistema multiple-input multiple-output (MIMO) con canales sin memoria. Para el escenario SISO se proponen tres diseños diferentes para un igualador probabilístico, tanto simple como turbo, que está basado en el algoritmo EP: una versión bloque, una versión filtrada que emula el comportamiento de un filtroWiener y una versión smoothing que funciona de forma similar a un Kalman smoother. Finalmente, la implementación del EP en bloque se adapta también para escenarios MIMO con realimentación desde el decodificador. En ambos escenarios, el EP se aplica de forma iterativa, incluyendo un mecanismo de damping y un control para evitar valores de varianzas negativas, que darían lugar a inestabilidades (especialmente, en constelaciones de alto orden). Los resultados experimentales que se incluyen en la tesis muestran que, cuando el algoritmo EP se aplica a sistemas de comunicaciones, se mejora notablemente el resultado de otras propuestas anteriores que existen en la literatura, con un pequeño incremento de la complejidad que es proporcional a la carga del LMMSE. Estos resultados también demuestran la robustez del algoritmo incluso para modulaciones de alto orden, canales con bastante memoria y un gran número de antenas.     A Low-Complexity Double EP-based Detector for Iterative Detection and Decoding in MIMO

    We propose a new iterative detection and decoding (IDD) algorithm for multiple-input multiple-output (MIMO) based on expectation propagation (EP) with application to massive MIMO scenarios. Two main results are presented. We first introduce EP to iteratively improve the Gaussian approximations of both the estimation of the posterior by the MIMO detector and the soft output of the channel decoder. With this novel approach, denoted by double-EP (DEP), the convergence is very much improved with a computational complexity just two times the one of the linear minimum mean square error (LMMSE) based IDD, as illustrated by the included experiments. Besides, as in the LMMSE MIMO detector, when the number of antennas increases, the computational cost of the matrix inversion operation required by the DEP becomes unaffordable. In this work we also develop approaches of DEP where the mean and the covariance matrix of the posterior are approximated by using the Gauss-Seidel and Neumann series methods, respectively. This low-complexity DEP detector has quadratic complexity in the number of antennas, as the low-complexity LMMSE techniques.     Structural basis of mitochondrial dysfunction in response to cytochrome c phosphorylation at tyrosine 48

    Regulation of mitochondrial activity allows cells to adapt to changing conditions and to control oxidative stress, and its dysfunction can lead to hypoxia-dependent pathologies such as ischemia and cancer. Although cytochrome c phosphorylation—in particular, at tyrosine 48—is a key modulator of mitochondrial signaling, its action and molecular basis remain unknown. Here we mimic phosphorylation of cytochrome c by replacing tyrosine 48 with p-carboxy-methyl-L-phenylalanine (pCMF). The NMR structure of the resulting mutant reveals significant conformational shifts and enhanced dynamics around pCMF that could explain changes observed in its functionality: The phosphomimetic mutation impairs cytochrome c diffusion between respiratory complexes, enhances hemeprotein peroxidase and reactive oxygen species scavenging activities, and hinders caspase-dependent apoptosis.     Balancing groundwater conservation and rural livelihoods under water and climate uncertainties: An integrated hydro-economic modeling framework

    In arid countries worldwide, social conflicts between irrigation-based human development and the conservation of aquatic ecosystems are widespread and attract many public debates. This research focuses on the analysis of water and agricultural policies aimed at conserving groundwater resources and maintaining rurallivelihoods in a basin in Spain's central arid region. Intensive groundwater mining for irrigation has caused overexploitation of the basin's large aquifer, the degradation of reputed wetlands and has given rise to notable social conflicts over the years. With the aim of tackling the multifaceted socio-ecological interactions of complex water systems, the methodology used in this study consists in a novel integration into a common platform of an economic optimization model and a hydrology model WEAP (Water Evaluation And Planning system). This robust tool is used to analyze the spatial and temporal effects of different water and agricultural policies under different climate scenarios. It permits the prediction of different climate and policy outcomes across farm types (water stress impacts and adaptation), at basin's level (aquifer recovery), and along the policies’ implementation horizon (short and long run). Results show that the region's current quota-based water policies may contribute to reduce water consumption in the farms but will not be able to recover the aquifer and will inflict income losses to the rural communities. This situation would worsen in case of drought. Economies of scale and technology are evidenced as larger farms with cropping diversification and those equipped with modern irrigation will better adapt to water stress conditions. However, the long-term sustainability of the aquifer and the maintenance of rurallivelihoods will be attained only if additional policy measures are put in place such as the control of illegal abstractions and the establishing of a water bank. Within the policy domain, the research contributes to the new sustainable development strategy of the EU by concluding that, in water-scarce regions, effective integration of water and agricultural policies is essential for achieving the water protection objectives of the EU policies. Therefore, the design and enforcement of well-balanced region-specific polices is a major task faced by policy makers for achieving successful water management that will ensure nature protection and human development at tolerable social costs. From a methodological perspective, this research initiative contributes to better address hydrological questions as well as economic and social issues in complex water and human systems. Its integrated vision provides a valuable illustration to inform water policy and management decisions within contexts of water-related conflicts worldwide

    Albiglutide and cardiovascular outcomes in patients with type 2 diabetes and cardiovascular disease (Harmony Outcomes): a double-blind, randomised placebo-controlled trial

    Background: Glucagon-like peptide 1 receptor agonists differ in chemical structure, duration of action, and in their effects on clinical outcomes. The cardiovascular effects of once-weekly albiglutide in type 2 diabetes are unknown. We aimed to determine the safety and efficacy of albiglutide in preventing cardiovascular death, myocardial infarction, or stroke. Methods: We did a double-blind, randomised, placebo-controlled trial in 610 sites across 28 countries. We randomly assigned patients aged 40 years and older with type 2 diabetes and cardiovascular disease (at a 1:1 ratio) to groups that either received a subcutaneous injection of albiglutide (30–50 mg, based on glycaemic response and tolerability) or of a matched volume of placebo once a week, in addition to their standard care. Investigators used an interactive voice or web response system to obtain treatment assignment, and patients and all study investigators were masked to their treatment allocation. We hypothesised that albiglutide would be non-inferior to placebo for the primary outcome of the first occurrence of cardiovascular death, myocardial infarction, or stroke, which was assessed in the intention-to-treat population. If non-inferiority was confirmed by an upper limit of the 95% CI for a hazard ratio of less than 1·30, closed testing for superiority was prespecified. This study is registered with ClinicalTrials.gov, number NCT02465515. Findings: Patients were screened between July 1, 2015, and Nov 24, 2016. 10 793 patients were screened and 9463 participants were enrolled and randomly assigned to groups: 4731 patients were assigned to receive albiglutide and 4732 patients to receive placebo. On Nov 8, 2017, it was determined that 611 primary endpoints and a median follow-up of at least 1·5 years had accrued, and participants returned for a final visit and discontinuation from study treatment; the last patient visit was on March 12, 2018. These 9463 patients, the intention-to-treat population, were evaluated for a median duration of 1·6 years and were assessed for the primary outcome. The primary composite outcome occurred in 338 (7%) of 4731 patients at an incidence rate of 4·6 events per 100 person-years in the albiglutide group and in 428 (9%) of 4732 patients at an incidence rate of 5·9 events per 100 person-years in the placebo group (hazard ratio 0·78, 95% CI 0·68–0·90), which indicated that albiglutide was superior to placebo (p<0·0001 for non-inferiority; p=0·0006 for superiority). The incidence of acute pancreatitis (ten patients in the albiglutide group and seven patients in the placebo group), pancreatic cancer (six patients in the albiglutide group and five patients in the placebo group), medullary thyroid carcinoma (zero patients in both groups), and other serious adverse events did not differ between the two groups. There were three (<1%) deaths in the placebo group that were assessed by investigators, who were masked to study drug assignment, to be treatment-related and two (<1%) deaths in the albiglutide group. Interpretation: In patients with type 2 diabetes and cardiovascular disease, albiglutide was superior to placebo with respect to major adverse cardiovascular events. Evidence-based glucagon-like peptide 1 receptor agonists should therefore be considered as part of a comprehensive strategy to reduce the risk of cardiovascular events in patients with type 2 diabetes.     Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Abstract Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    Revisión de algoritmos de detección para sistemas MIMO de alto orden

    Con este trabajo se pretende comparar varios algoritmos de detección para sistemas de comunicación MIMO y constelaciones de gran orden, dado que la utilización del detector óptimo de máxima verosimilitud (ML) es inabordable computacionalmente. Para este fin, se han creado varias funciones en código matlab que se han publicado como toolbox en mathworks: http://www.mathworks.com/matlabcentral/fileexchange/48212-detection-mimo-toolbox Entre los algoritmos de detección destacan el ZF, MMSE y sus versiones con cancelación de interferencias (SIC), las cuales proporcionan una mejora de la probabilidad de error a cambio de una mayor carga computacional. Sin embargo, este trabajo se centra tanto en el algoritmo GTA [1], [2] como en el algoritmo EP [13] (publicada recientemente su aplicación a los sistemas MIMO). - El algoritmo GTA (aproximación de árbol gaussiana) se basa en aproximar la distribución de probabilidad a posteriori gaussiana exacta mediante un árbol libre de bucles. A continuación, se aplica la restricción de que los símbolos pertenecen a un conjunto finito de datos (determinados por el tipo y tamaño de la constelación) para obtener una aproximación de la distribución de probabilidad discreta libre de bucles, la cual se maximiza mediante un algoritmo de paso de mensajes, i.e, el algoritmo BP. - El algoritmo EP (expectation propagation) está basado en buscar una aproximación a la distribución de probabilidad a posteriori a través de una distribución gaussiana cuya media y varianza estén lo más próximas posible a la media y la varianza de la distribución exacta original. Esto se consigue a través de un método iterativo. Los resultados de simulación muestran que la probabilidad de error del algoritmo GTA (en concreto, su versión con cancelación de interfencias) y de EP, son bastante mejores si se comparan con los algoritmos ZF y MMSE, tanto en su versión estándar como SIC

    Jornadas Nacionales de Robótica y Bioingeniería 2023: Libro de actas

    Las Jornadas de Robótica y Bioingeniería de 2023 tienen lugar en la Escuela Técnica Superior de Ingeniería Industrial de la Universidad Politécnica de IVIadrid, entre los días 14 y 16 de junio de 2023. En este evento propiciado por el Comité Español de Automática (CEA) tiene lugar la celebración conjunta de las XII Jornadas Nacionales de Robótica y el XIV Simposio CEA de Bioingeniería. Las Jornadas Nacionales de Robótica es un evento promovido por el Grupo Temático de Robótica (GTRob) de CEA para dar visibilidad y mostrar las actividades desarrolladas en el ámbito de la investigación y transferencia tecnológica en robótica. Asimismo, el propósito de Simposio de Bioingeniería, que cumple ahora su decimocuarta dicción, es el de proporcionar un espacio de encuentro entre investigadores, desabolladores, personal clínico, alumnos, industriales, profesionales en general e incluso usuarios que realicen su actividad en el ámbito de la bioingeniería. Estos eventos se han celebrado de forma conjunta en la anualidad 2023. Esto ha permitido aunar y congregar un elevado número de participantes tanto de la temática robótica como de bioingeniería (investigadores, profesores, desabolladores y profesionales en general), que ha posibilitado establecer puntos de encuentro, sinergias y colaboraciones entre ambos. El programa de las jornadas aúna comunicaciones científicas de los últimos resultados de investigación obtenidos, por los grupos a nivel español más representativos dentro de la temática de robótica y bioingeniería, así como mesas redondas y conferencias en las que se debatirán los temas de mayor interés en la actualidad. En relación con las comunicaciones científicas presentadas al evento, se ha recibido un total de 46 ponencias, lo que sin duda alguna refleja el alto interés de la comunidad científica en las Jornadas de Robótica y Bioingeniería. Estos trabajos serán expuestos y presentados a lo largo de un total de 10 sesiones, distribuidas durante los diferentes días de las Jornadas. Las temáticas de los trabajos cubren los principales retos científicos relacionados con la robótica y la bioingeniería: robótica aérea, submarina, terrestre, percepción del entorno, manipulación, robótica social, robótica médica, teleoperación, procesamiento de señales biológicos, neurorehabilitación etc. Confiamos, y estamos seguros de ello, que el desarrollo de las jornadas sea completamente productivo no solo para los participantes en las Jornadas que podrán establecer nuevos lazos y relaciones fructíferas entre los diferentes grupos, sino también aquellos investigadores que no hayan podido asistir. Este documento que integra y recoge todas las comunicaciones científicas permitirá un análisis más detallado de cada una de las mismas
